1.Transgenic Fish Research
Faster-growing transgenic fish: The “all-fish” growth hormone gene construct was transferred into the fertilized eggs of common Carp, and fast-growing transgenic fish were successfully produced. The growth rate and feed conversion efficiency of F1 transgenic common carp are 42% and 11% higher over the control respectively.
Disease resistant transgenic fish: Using outer capsid protein VP7 gene of grass carp reovirus (GCRV) as target gene, and employing grass carp H1 promoter, a shRNA expression construct was constructed. Transferred this construct into the fertilized eggs of rare minnow, and obtained the GCRV resistant transgenic rare minnow.
2. Fish Genomics and Molecular Breeding Research
Construction of Libraries: Two BAC libraries of grass carp in the EcoRI and HindIII sites of the vector CopyControl pCC1BAC were constructed, providing a totally genome coverage of 14.2× haploid genome equivalents of grass carp. Two cDNA libraries from grass carp head kidney and spleen were also constructed.
EST sequencing: single pass sequencing on 10,000 randomly selected clones from grass carp head kidney cDNA library was performed, and 3074 unigenes were obtained.
Development of molecular markers: Over 1,000 SSR markers of grass carp were cloned, and SNP markers from immune-related genes were detected.
Linkage mapping and QTL mapping: In present, I am working on genetic linkage mapping of grass carp with SSR and SNP markers, and QTL mapping on growth rate, feed conversion efficiency and GCRV resistant traits.
